Elenagen

Elenagen is a novel DNA immunotherapy and experimental biologic agent consisting of a supercoiled circular DNA (plasmid) encoding the human gene p62/SQSTM1. Developed by CureLab Oncology, it was initially designed as an anti-cancer DNA vaccine to elicit an adaptive immune response against tumor-associated antigens highly expressed in cancer cells. Its mechanism of action is multifaceted: - It modifies the tumor microenvironment to enable immune cell infiltration into tumors, addressing resistance seen in "cold" tumors. - It stimulates both innate and adaptive anti-tumor immunity. - It indirectly modulates mesenchymal stem cells (MSCs), enhancing their production of trophic and anti-inflammatory factors even without direct exposure—this effect may contribute to tissue repair and systemic reduction of chronic inflammation. - Preclinical studies show additional benefits such as reversal or prevention of osteoporosis via rejuvenation of MSCs' osteogenic potential. Clinically, Elenagen has demonstrated significant benefit when combined with chemotherapy (e.g., gemcitabine) for platinum-resistant ovarian cancer patients by prolonging progression-free survival. Beyond oncology, it shows promise for chronic inflammatory diseases (e.g., psoriasis), diabetic foot ulcers, neurodegenerative diseases (ALS, Alzheimer’s disease, Huntington’s disease, Parkinson’s disease), and age-related conditions due to its broad immunomodulatory effects[1][2][4][5][7][8][9][10].
