Overview

Elgemtumab is a fully human IgG1κ monoclonal antibody that targets the receptor tyrosine-protein kinase erbB-3 (HER3), a member of the ERBB family of receptor tyrosine kinases. By binding to HER3, elgemtumab blocks both ligand-dependent and independent HER3 signal transduction and cell proliferation. This inhibition disrupts downstream signaling pathways such as PI3K/AKT and prevents tumor growth in models overexpressing HER2 or neuregulin (NRG). Elgemtumab was developed for its anti-tumor potential in cancers where HER3-mediated signaling contributes to oncogenic transformation. It has shown synergistic effects when combined with other agents like trastuzumab or PI3K inhibitors. The drug was originally developed by MorphoSys and Novartis Oncology, with clinical development discontinued for indications including breast cancer, gastric cancer, head and neck cancer, esophageal cancer, and solid tumors[1][2][3][4][5][8].
