**ELI-002 2P** is an investigational, off-the-shelf lymph node-targeted therapeutic cancer vaccine developed by Elicio Therapeutics for minimal residual disease-positive, KRAS-mutated pancreatic ductal adenocarcinoma and colorectal cancer after locoregional therapy. It contains amphiphile-modified long peptides encoding the KRAS G12D and G12R mutations plus the amphiphile-modified Toll-like receptor 9 agonist CpG-7909. The lipid-like amphiphile modifications bind endogenous albumin after subcutaneous injection and promote lymphatic transport to draining lymph nodes, where antigen-presenting cells stimulate mutant-KRAS-specific CD4-positive and CD8-positive T-cell responses. In the completed phase 1 AMPLIFY-201 study, ELI-002 2P showed an acceptable safety profile and induced mutant-KRAS-specific T-cell responses and reductions in circulating tumor biomarkers; it is not approved for clinical use.
