ELI-004 is an AMP-modified CpG oligodeoxynucleotide adjuvant developed using Elicio Therapeutics’ proprietary Amphiphile (AMP) technology. It is designed to enhance immune responses by targeting lymph nodes, where it stimulates antigen-presenting cells such as dendritic cells to activate T and B cell-mediated immunity. The AMP modification enables efficient delivery of the CpG oligonucleotide to lymph nodes via albumin binding, increasing its immunostimulatory potency. ELI-004 is used as an off-the-shelf subcutaneous adjuvant component in investigational cancer vaccines (notably in combination with mutant KRAS peptide antigens in the ELI-002 vaccine) and has also been studied as a vaccine adjuvant for infectious diseases such as COVID-19[1][2][3][4][5][7][9].
