ELI-008

ELI-008 is an investigational multivalent lymph node–targeted AMP peptide vaccine developed by Elicio Therapeutics. It is designed to target p53 hotspot mutations, which are among the most common genetic alterations in human solid tumors. The vaccine uses Elicio’s proprietary Amphiphile (AMP) platform to deliver modified mutant p53 peptide antigens directly to the lymph nodes, enhancing immune system engagement and T cell activation against tumor cells harboring these mutations. Preclinical studies have demonstrated that ELI-008 induces robust and highly potent tumor antigen-specific T cell responses—often several-hundred-fold higher than conventional vaccines—against a range of common p53 mutations (including R248W, R248Q, R175H, R273H, R273C, R282W, G245S, Y220C, C135Y, R158H, H214R). The AMP technology works by latching onto albumin at the injection site for efficient delivery to lymphatic tissue. Mutations in p53 are present in over 30% of solid tumors and lead to uncontrolled tumor progression; thus ELI-008 aims for broad applicability across multiple cancer types[2][5][9].
