Overview

ELI-216 is an investigational cancer vaccine developed by Elicio Therapeutics, utilizing the proprietary Amphiphile (AMP) platform to target malignancies driven by the BRAF V600E mutation. The vaccine is designed to deliver mutation-specific peptide antigens and a potent adjuvant, AMP-CpG, directly to the lymph nodes, which are the primary sites for coordinating immune responses. By mimicking the natural transport of fatty acids bound to albumin, the AMP platform ensures efficient delivery to the lymphatic system, where it activates a robust and durable response from both CD4+ and CD8+ T cells. This targeted approach aims to overcome the limitations of traditional peptide vaccines, which often fail to reach the lymph nodes effectively, and is intended for the treatment of BRAF-mutant cancers such as melanoma, colorectal cancer, and non-small cell lung cancer.
