Elinzanetant is an investigational, oral, once-daily **small-molecule dual neurokinin receptor antagonist** being developed by Bayer as a **non-hormonal treatment for moderate to severe vasomotor symptoms**. It antagonizes **neurokinin 1 and neurokinin 3 receptors**, with the therapeutic rationale of modulating estrogen-sensitive **KNDy neurons** in the hypothalamus and thereby reducing the thermoregulatory dysfunction that drives hot flashes. The most advanced development program is the global Phase III **OASIS** program, including studies in vasomotor symptoms associated with menopause and in vasomotor symptoms caused by adjuvant endocrine therapy for hormone receptor-positive breast cancer or breast cancer risk reduction. Publicly reported data show positive Phase III results in OASIS 1, 2, 3, and 4, including statistically significant reductions in hot-flash frequency and severity, along with improvements in sleep disturbance and menopause-related quality of life.
