Drug intelligence / Profile preview
Elisrasib plus cetuximab is an investigational combination regimen being evaluated for the treatment of KRAS G12C-mutant solid tumors, particularly colorectal cancer (CRC). Elisrasib (D3S-001) is an orally bioavailable, next-generation covalent small-molecule inhibitor that selectively targets the inactive GDP-bound state of mutant KRAS G12C, designed for rapid and complete target engagement. Cetuximab (Erbitux) is an intravenously administered chimeric monoclonal antibody targeting the epidermal growth factor receptor (EGFR). In KRAS G12C-mutant colorectal cancer, EGFR-mediated reactivation of the MAPK pathway is a primary mechanism of resistance to KRAS G12C inhibitors. Combining elisrasib with cetuximab is designed to vertically inhibit this pathway, preventing EGFR-driven reactivation and deepening therapeutic responses. The combination is being developed by D3 Bio and is currently in Phase 1/2 clinical trials.
