Overview

**Elliptinium** is a semi-synthetic derivative of ellipticine, an alkaloid originally isolated from the Apocynaceae plant family. It acts as an antineoplastic agent by intercalating into DNA, thereby inhibiting DNA replication and RNA/protein synthesis. The drug primarily exerts its effect by stabilizing the cleavable complex formed by topoisomerase II, leading to DNA breaks[3]. Elliptinium has demonstrated modest clinical activity as salvage therapy in advanced breast cancer and other solid tumors, though its use is limited due to significant toxicities, including xerostomia and, with certain regimens, immune-mediated hemolytic reactions due to anti-elliptinium IgM antibodies[2]. It is marketed under the brand name Celiptium, and the most common pharmaceutical salt is elliptinium acetate[1][2][3][4].
