Overview

Elmustine (HECNU) is a water-soluble small molecule chemotherapeutic agent belonging to the nitrosourea class of alkylating agents. It was developed by ASTA Medica, Faustus Forschung Compagnie, and Translational Cancer Research GmbH for the treatment of various malignancies, including progressive pancreatic cancer and malignant gliomas. Elmustine's mechanism of action involves DNA alkylation, specifically inducing DNA-DNA interstrand crosslinking, which disrupts DNA replication and transcription, leading to cell cycle arrest and apoptosis. Although it demonstrated clinical activity in Phase II trials for glioma and pancreatic cancer, its development has been discontinued.
