ELND-001 is an orally bioavailable small molecule gamma-secretase inhibitor (GSI) that was under development by Elan Corporation for the treatment of Alzheimer's disease. The drug was designed to modulate the amyloidogenic pathway by inhibiting the gamma-secretase enzyme complex, which is responsible for the final proteolytic cleavage of amyloid precursor protein (APP) into amyloid-beta (Aβ) peptides. By reducing the generation of neurotoxic Aβ isoforms, particularly Aβ42, ELND-001 aimed to prevent the formation of amyloid plaques and slow the progression of cognitive decline. Development of ELND-001 was discontinued during early-stage clinical trials. This cessation was largely influenced by the broader clinical challenges faced by the GSI class, including the high-profile failure of semagacestat and concerns regarding off-target inhibition of Notch signaling, which is associated with gastrointestinal and immunological toxicities.
