Overview

ELND002 is a pegylated small molecule integrin alpha4 (ITGA4) inhibitor that was developed as an injectable therapy for the treatment of multiple sclerosis and hematologic malignancies. Its mechanism of action involves antagonism of integrin alpha4 (also known as CD49d), which plays a key role in leukocyte adhesion and migration across the blood-brain barrier—a process implicated in the pathogenesis of multiple sclerosis. The drug was originally developed by Elan Corporation and later associated with Perrigo. Clinical development reached Phase 1 trials for relapsing forms of secondary progressive multiple sclerosis (SPMS) and relapsing-remitting multiple sclerosis (RRMS), but all studies were terminated or discontinued before further advancement[1][2][3][5].
