Overview

**Elotuzumab + bortezomib + dexamethasone** is a three-drug combination regimen used in the treatment of relapsed or refractory multiple myeloma. **Elotuzumab** is a humanized monoclonal antibody that targets the Signaling Lymphocytic Activation Molecule F7 (SLAMF7), activating natural killer (NK) cells and facilitating antibody-dependent cell-mediated cytotoxicity against myeloma cells. **Bortezomib** is a small molecule proteasome inhibitor that blocks the 26S proteasome, leading to apoptosis of cancer cells. **Dexamethasone** is a synthetic glucocorticoid corticosteroid that exerts anti-inflammatory and immunosuppressive effects. This regimen is designed to leverage direct cytotoxic effects (bortezomib, dexamethasone) and immune-mediated mechanisms (elotuzumab) for improved disease control. Primary indication: relapsed/refractory multiple myeloma[4][6].
