Overview

elotuzumab + lenalidomide + autologous stem cell transplantation is a combination therapeutic regimen investigated for the treatment of symptomatic multiple myeloma. This regimen integrates elotuzumab (Empliciti), a humanized monoclonal antibody targeting SLAM family member 7 (SLAMF7), with lenalidomide (Revlimid), an immunomodulatory imide drug (IMiD) that binds to cereblon to induce the degradation of transcription factors Ikaros and Aiolos. The protocol involves induction therapy, followed by high-dose chemotherapy (typically melphalan) and autologous stem cell transplantation (ASCT), with subsequent maintenance therapy using elotuzumab and lenalidomide. The combination aims to leverage the direct activation of natural killer cells and the tagging of myeloma cells by elotuzumab alongside the tumor-inhibiting and immune-modulating effects of lenalidomide to improve outcomes following transplant.
