Elotuzumab + nivolumab is an experimental combination therapy consisting of two monoclonal antibodies with distinct mechanisms of action, studied primarily for the treatment of relapsed/refractory multiple myeloma. Elotuzumab targets SLAMF7 (signaling lymphocytic activation molecule F7), activating natural killer (NK) cells and mediating antibody-dependent cellular cytotoxicity against myeloma cells, while also facilitating immune modulation through Fc receptor engagement. Nivolumab is a programmed death-1 (PD-1) immune checkpoint inhibitor blocking PD-1/PD-L1 interaction, thereby restoring T-cell activity and promoting antitumor immunity. The combination was investigated in clinical trials such as CheckMate 602, with the hypothesis that PD-1 pathway blockade may potentiate elotuzumab’s immunomodulatory effect; however, no significant improvement in efficacy over standard therapy was demonstrated in phase 3 studies[1][7].
