This combination regimen consists of **elotuzumab** (a humanized monoclonal antibody targeting signalling lymphocytic activation molecule family member 7), **thalidomide** (an immunomodulatory agent), **dexamethasone** (a synthetic glucocorticoid corticosteroid), and **cyclophosphamide** (an alkylating agent). Used experimentally for relapsed/refractory multiple myeloma, this combination seeks to improve antitumor activity through multiple mechanisms: elotuzumab stimulates natural killer cells and mediates antibody-dependent cellular cytotoxicity of myeloma cells; thalidomide suppresses angiogenesis and modulates immune response; dexamethasone provides anti-inflammatory and cytotoxic effects; cyclophosphamide causes DNA damage in rapidly dividing cells. In Phase II trials, the regimen was well tolerated and showed moderate efficacy in heavily pre-treated multiple myeloma patients[1][2][5].
