Overview

ELP-p21 is a thermally responsive macromolecular conjugate consisting of an elastin-like polypeptide (ELP) carrier fused to a p21-derived therapeutic peptide. The ELP component provides a phase-transition property, allowing the construct to remain soluble at physiological temperatures but aggregate and accumulate in tissues subjected to local hyperthermia (above 40°C). The p21 peptide component is designed to inhibit the cell cycle at the G1/S transition by mimicking the activity of the endogenous cyclin-dependent kinase inhibitor p21 (WAF1/CIP1). Unlike its counterpart Bac-ELP-p21, ELP-p21 lacks a cell-penetrating peptide (CPP), relying on passive targeting and thermal accumulation for tumor uptake. It has been investigated primarily in preclinical models of pancreatic cancer.
