ELP63-MMP-2i is a kidney-targeted recombinant fusion protein therapeutic designed for the treatment of renal fibrosis and chronic kidney disease. It consists of a selective peptide inhibitor of matrix metalloproteinase-2 (MMP-2i) fused to a 63-repeat elastin-like polypeptide (ELP) scaffold. The ELP component serves as a macromolecular drug delivery system that facilitates renal accumulation and extends the therapeutic's plasma half-life. By selectively inhibiting MMP-2, the drug aims to restore extracellular matrix homeostasis and prevent the pathological remodeling associated with hypertensive renal injury. Preclinical studies in Dahl salt-sensitive rats have demonstrated that ELP63-MMP-2i treatment significantly reduces proteinuria, albuminuria, and renal collagen deposition compared to vehicle controls.
