ELP77 is a recombinant, thermally responsive elastin-like polypeptide (ELP) composed of 77 pentapeptide repeats (Val-Pro-Gly-Xaa-Gly) with valine, glycine, and alanine as guest residues. Developed as a biocompatible and biodegradable macromolecular carrier, it undergoes an inverse temperature phase transition, shifting from a soluble state to insoluble aggregates at temperatures above its transition temperature (approximately 38.67°C). In oncological research, specifically regarding bladder cancer, ELP77 is utilized as a non-targeted control polymer to establish a baseline for passive accumulation via the enhanced permeability and retention (EPR) effect, allowing researchers to evaluate the added efficacy of ligand-modified variants like B5V60. Its molecular weight is approximately 31.3 kDa, and it is designed to remain soluble at physiological temperatures while providing a versatile scaffold for drug delivery applications.
