Overview

Elpamitide is a synthetic peptide vaccine consisting of a 9-amino acid sequence (RFVPDGNRI) derived from the vascular endothelial growth factor receptor 2 (VEGFR2). It is designed to induce a cytotoxic T-lymphocyte (CTL) response against VEGFR2-expressing endothelial cells within the tumor neovasculature. By targeting the tumor's blood supply rather than the tumor cells directly, elpamitide aims to inhibit angiogenesis and suppress tumor growth. It has been primarily investigated in clinical trials for advanced pancreatic and gastric cancers, often in combination with chemotherapy such as gemcitabine. The vaccine is specifically restricted to patients with the HLA-A*2402 genotype, as the peptide epitope is presented by this specific human leukocyte antigen.
