**ELT048** is a first-in-class antibody-drug conjugate (ADC) targeting L1 cell adhesion molecule (L1CAM), a cell surface glycoprotein overexpressed in pancreatic and ovarian cancers that promotes tumor cell proliferation and migration. Developed from a proprietary humanized mouse monoclonal antibody conjugated to a cytotoxic payload via LegoChem's linker technology, it demonstrated excellent anti-tumor efficacy in preclinical in vitro and in vivo models, good biophysical and pharmacokinetic properties, and an acceptable therapeutic index. In exploratory GLP toxicology studies in cynomolgus monkeys, ELT048 was well-tolerated with no treatment-related macroscopic or microscopic changes observed, confirming exposure and pharmacodynamic activity.
