Overview

Eltrombopag + adagrasib is a combination of two small-molecule drugs, each with distinct mechanisms and indications. Eltrombopag is a thrombopoietin receptor agonist that stimulates the production of platelets by activating the thrombopoietin receptor (TPO-R) on bone marrow cells. It is primarily used to treat chronic immune thrombocytopenia (ITP), severe aplastic anemia, and to increase platelet counts in patients with hepatitis C undergoing interferon-based therapy[1][5][6]. Adagrasib is a targeted kinase inhibitor that selectively inhibits KRAS G12C, an oncogenic driver mutation found in certain cancers. By blocking KRAS G12C activity, adagrasib disrupts cancer cell proliferation and survival. It is approved for use in non-small cell lung cancer (NSCLC) with KRAS G12C mutations after prior therapy, and in combination with cetuximab for metastatic colorectal cancer harboring the same mutation[3][4][7][8]. This combination does not represent an established therapeutic regimen but rather lists both agents together.
