**Elunetirom (ABX-002)** is an orally administered, potent, selective, CNS-directed **thyroid hormone receptor beta (THR-β or TRβ)** agonist prodrug developed by **Autobahn Therapeutics** primarily as an adjunctive therapy for **major depressive disorder (MDD)** and **bipolar disorder depression**. It penetrates the blood-brain barrier and is cleaved by fatty acid amide hydrolase (FAAH) to its active metabolite (ABX-002A), which activates TRβ to promote oligodendrocyte differentiation, myelination/remyelination, brain bioenergetics, and potentiation of serotonergic antidepressant effects while minimizing peripheral side effects like cardiac toxicity. A **small molecule** with molecular formula **C19H21Cl2NO3**, it has completed Phase 1 trials showing dose-proportional pharmacokinetics and is currently in **Phase 2** for MDD and bipolar depression, with orphan drug designation for **X-linked adrenoleukodystrophy** and preclinical data in multiple sclerosis.
