Overview

This drug is a **combination regimen** consisting of ELVN-002, trastuzumab, 5-fluorouracil, leucovorin, and oxaliplatin. - **ELVN-002** is an irreversible and CNS-penetrant selective HER2 tyrosine kinase inhibitor designed to target wild-type and mutant HER2, sparing wild-type EGFR, and is under development for HER2-positive and HER2-mutant solid tumors, including breast and non-small cell lung cancer[3][5]. - **Trastuzumab** is a monoclonal antibody targeting the extracellular domain of the HER2 (human epidermal growth factor receptor 2), blocking HER2-mediated signaling and mediating antibody-dependent cellular cytotoxicity in HER2-positive cancers[2][4][6]. - **5-Fluorouracil** is a small molecule antimetabolite that interferes with DNA synthesis by inhibiting thymidylate synthase, exerting cytotoxic effects primarily during the S phase of the cell cycle. - **Leucovorin** (folinic acid) is used to enhance the efficacy and reduce the toxicity of 5-fluorouracil by stabilizing the 5-FU-thymidylate synthase complex. - **Oxaliplatin** is a platinum-based alkylating agent that forms DNA crosslinks and inhibits DNA replication. This combination is intended for the treatment of HER2-positive and HER2-mutant solid tumors. ELVN-002’s mechanism and approval status remain investigational (early clinical trials)[3][5]; the entire combination has not received regulatory approval and is being tested in clinical studies.
