ELVN-002 specifically inhibits wild-type and mutant HER2 kinases with minimal off-target effects on EGFR, aiming to address resistance, brain metastases, and toxicity issues in HER2-positive and HER2-mutant cancers[1][2][4][7]. Trastuzumab targets the extracellular domain of HER2, blocking receptor activation and downstream signaling, while paclitaxel disrupts microtubule dynamics, leading to apoptotic cell death. This combination is being explored for synergistic antitumor activity in cancers with HER2 alterations, including metastatic breast cancer and non-small cell lung cancer, particularly in post-ADC and post-anti-HER2 therapy settings[1][4][7].
