Overview

**EM1032** is a novel tetravalent 2+2 T-cell engager (TCE) bispecific antibody developed using EpimAb Biotherapeutics' proprietary Fabs-In-Tandem Immunoglobulin (FIT-Ig) platform with binding domains in cis-configuration. It simultaneously targets **ALPP** (alkaline phosphatase, placental type) and/or **ALPG** (alkaline phosphatase, germ cell type) on tumor cells and **CD3** on T cells, recruiting and activating T cells to induce potent tumor cell lysis specifically in the presence of ALPP+ target cells. Preclinical studies demonstrate hundreds-fold greater cytotoxicity than ALPP-targeting antibody-drug conjugates (ADCs) like SGN-ALPV analogue and h12F3-DXd, eradication of ovarian cancer cell line-derived xenografts (CDX) resistant to ADCs, increased tumor-infiltrated CD8+ and CD4+ T cells, and favorable cytokine release profiles. It shows promise for ALPP(G)-expressing solid tumors, particularly platinum- and ADC-resistant ovarian cancer, with IND-enabling studies ongoing.[1][2][3]
