EM1033 is a protease-activated bispecific T-cell engager (TCE) developed by EpimAb Biotherapeutics for the treatment of solid tumors. Utilizing EpimAb's proprietary ProTCE platform, EM1033 is designed to target CD228 (melanoma-associated antigen p97) and CD3. The molecule is engineered to remain in an inactive state in systemic circulation and undergo conditional activation by proteases specifically found within the tumor microenvironment. This approach aims to broaden the therapeutic window by reducing off-tumor, on-target toxicities while facilitating potent T-cell mediated destruction of CD228-expressing cancer cells. As of 2024, the program is in the IND-enabling/preclinical stage of development.
