Overview

EM164 is a murine monoclonal antibody that targets the type 1 insulin-like growth factor receptor (IGF-1R, also known as CD221). Developed by ImmunoGen, EM164 binds specifically and with high affinity (Kd ~0.1 nM) to the extracellular domain of human IGF-1R, blocking the binding of its ligands, IGF-1 and IGF-2, and preventing receptor autophosphorylation and downstream signaling through the PI3K/Akt and MAPK pathways. It also induces receptor internalization and degradation. EM164 served as the precursor for the humanized version, AVE1642 (also known as VRDN-001 or veligrotug). Veligrotug was evaluated in clinical trials for oncology indications by Sanofi-Aventis and was subsequently licensed to Viridian Therapeutics, which successfully developed it for thyroid eye disease (TED). Veligrotug (under the brand name Lumvoa) was approved by the FDA on June 26, 2026, for the treatment of active and chronic thyroid eye disease.
