Emactuzumab is a humanized monoclonal antibody that targets the colony stimulating factor 1 receptor (CSF1R, also known as CD115 or macrophage colony-stimulating factor receptor). By binding to CSF1R on macrophages, emactuzumab blocks the interaction between CSF-1 and its receptor, inhibiting downstream signaling. This action depletes tumor-associated macrophages (TAMs), which are implicated in immune suppression and tumor progression. The reduction of TAMs enhances T-cell infiltration and antitumor immune responses within the tumor microenvironment. Emactuzumab is being developed primarily for tenosynovial giant cell tumor (TGCT), especially in patients not amenable to surgery, but has also been investigated for other cancers such as advanced solid tumors and non-Hodgkin lymphoma. It has demonstrated high response rates and durable clinical benefit in TGCT trials with a manageable safety profile[1][7][8][10].
