Overview

Emactuzumab + RO7009789 is a combination of two monoclonal antibodies under investigation for cancer immunotherapy, particularly in advanced solid tumors. Emactuzumab targets **colony-stimulating factor 1 receptor (CSF1R)**, reducing tumor-associated macrophages (TAMs) to decrease immunosuppression in the tumor microenvironment. RO7009789 (also known as selicrelumab) is an agonistic antibody targeting **CD40**, a protein on antigen-presenting cells and some tumor cells; it stimulates immune activation, leading to apoptosis in CD40-positive cancer cells and enhancement of B and T cell immune responses. The combined rationale is to simultaneously inhibit the immunosuppressive TAMs and activate the adaptive immune response to increase anti-tumor efficacy. The combination has been studied in phase 1 clinical trials for advanced solid tumors[1][3][5][7][9].
