emapalumab

Emapalumab is a fully human monoclonal antibody that targets and neutralizes interferon-gamma (IFN-γ), a cytokine implicated in the pathogenesis of primary hemophagocytic lymphohistiocytosis (HLH). HLH is a rare, life-threatening hyperinflammatory syndrome characterized by excessive activation of T lymphocytes and macrophages, leading to severe immune-mediated tissue damage. Emapalumab binds with high affinity to both free and receptor-bound IFN-γ, inhibiting its interaction with the IFN-γ receptor on immune cells and thereby blocking downstream inflammatory signaling via the JAK-STAT pathway. This mechanism reduces pathological immune activation in patients with HLH who are refractory, recurrent, or intolerant to conventional therapies. Emapalumab was developed by Novimmune and is marketed under the brand name Gamifant.
