Drug intelligence / Profile preview
Emavusertib + azacitidine + venetoclax is an investigational triple-drug combination regimen being developed by Curis for the treatment of acute myeloid leukemia (AML). The regimen combines emavusertib (CA-4948), an oral small-molecule inhibitor of interleukin-1 receptor-associated kinase 4 (IRAK4) and FMS-like tyrosine kinase 3 (FLT3), with the standard-of-care backbone of azacitidine (a hypomethylating agent) and venetoclax (a BCL-2 inhibitor). This triplet therapy is designed to target multiple survival pathways in leukemia cells and overcome resistance mechanisms, such as FLT3 mutations or MCL-1 upregulation, which often lead to relapse on the doublet of azacitidine and venetoclax. It is currently being evaluated in clinical trials, including the Phase 1 TakeAim Leukemia study (CA-4948-104), as an add-on therapy to achieve undetectable measurable residual disease (uMRD) in AML patients who are in complete response but remain MRD-positive.
