Overview

EMB-15 (also known as ES015) is a bispecific T-cell engager (BiTE) antibody developed by Elpiscience Biopharmaceuticals using its proprietary FIT-Ig (Fabs-in-tandem immunoglobulin) platform. It is designed to simultaneously target Claudin 18.2 (CLDN18.2) on tumor cells and the CD3 epsilon subunit on T-cells. By cross-linking these two targets, EMB-15 facilitates the formation of an immunological synapse between T-cells and CLDN18.2-positive cancer cells, leading to T-cell activation and subsequent tumor cell lysis. CLDN18.2 is a highly specific tight junction protein frequently overexpressed in various solid tumors, particularly gastric, pancreatic, and esophageal cancers, while remaining largely absent in normal tissues. EMB-15 is currently being evaluated in Phase I clinical trials for patients with locally advanced or metastatic solid tumors who have exhausted standard treatment options.
