Embeda is an abuse-deterrent, extended-release oral formulation combining morphine sulfate, a mu-opioid receptor agonist, with a sequestered core of naltrexone hydrochloride, a mu-opioid receptor antagonist. Developed by Pfizer (following the acquisition of King Pharmaceuticals), it is indicated for the management of moderate-to-severe chronic pain requiring daily, around-the-clock, long-term opioid treatment. The formulation is specifically designed to deter misuse; when taken as directed, the naltrexone remains sequestered and exerts no clinical effect. However, if the pellets are crushed or chewed, the naltrexone is released and absorbed, potentially neutralizing the euphoric effects of morphine and precipitating withdrawal in opioid-dependent individuals.
