EMBL205 is a small molecule inhibitor of Muscle RING-finger protein-1 (MuRF1), an E3 ubiquitin ligase primarily expressed in cardiac and skeletal muscle. MuRF1 plays a critical role in the ubiquitin-proteasome pathway by mediating the degradation of sarcomeric proteins, which contributes to muscle wasting (atrophy) and cardiac remodeling. Developed by researchers at the Technische Universität Dresden, EMBL205 is being investigated for its potential to treat heart failure with preserved ejection fraction (HFpEF) and associated skeletal muscle dysfunction. In preclinical models, such as the ZSF1 rat, EMBL205 has demonstrated the ability to attenuate myocardial hypertrophy, reduce perivascular fibrosis, and improve diastolic function by lowering the expression of collagen (Col1A, Col3A) and matrix metalloproteinase-2 (MMP2).
