Embryonic stem cell-derived mesenchymal stem cells (ES-MSCs) are a specialized population of mesenchymal stem cells generated from human embryonic stem cell (hESC) lines. They are being investigated as a therapeutic platform for neurodegenerative diseases, most notably Alzheimer's disease (AD). Preclinical studies indicate that ES-MSCs can reduce the accumulation of amyloid-beta (Aβ) in the brain by enhancing autophagolysosomal clearance mechanisms. Additionally, these cells demonstrate superior viability and neuroprotective capacity compared to traditional bone marrow-derived MSCs (BM-MSCs) in amyloid-rich environments. Clinical interest in ES-MSCs stems from their potential for large-scale production and their ability to improve cognitive function and inhibit hippocampal cell death when administered via intra-arterial or other systemic routes.
