Overview

EMD 521873 + cyclophosphamide is a combination therapy using a recombinant fusion protein known as EMD 521873 (Selectikine, NHS-IL2-LT), which consists of a genetically modified human interleukin-2 (IL-2) fused to an antibody specific for necrotic DNA (NHS76), together with the alkylating agent cyclophosphamide. Selectikine targets high-affinity IL-2 receptors to stimulate T-cell activation and is designed to selectively concentrate IL-2 activity at tumor sites. Cyclophosphamide is added at a low dose to modulate immune response, particularly by reducing regulatory T-cell activity. The combination has been investigated in phase I trials for patients with metastatic or locally advanced solid tumors and B-cell non-Hodgkin lymphoma, focusing on safety, pharmacodynamics, and preliminary efficacy.
