Overview

EMD534085 is a potent and selective small molecule inhibitor of Eg5 (also known as Kinesin-like protein KIF11), a microtubule motor protein essential for the formation of a bipolar spindle during mitosis. By inhibiting Eg5, EMD534085 induces mitotic arrest characterized by the formation of monastral spindles, which subsequently leads to cell cycle arrest and apoptosis. Developed by Merck Serono, the compound was evaluated in a Phase I dose-escalation study in patients with advanced solid tumors and lymphomas. Clinical results indicated that the drug was generally well-tolerated, with dose-limiting toxicities primarily involving transient neutropenia and cardiac events. However, significant objective tumor responses were not observed in early clinical testing, and development of the asset appears to have been terminated.
