EMP108 is a targeted radioligand therapy (RLT) currently in preclinical development by Empiriko for the treatment of prostate-specific membrane antigen (PSMA)-expressing prostate cancer. The therapeutic complex consists of a PSMA-targeting antibody conjugated to the beta-emitting radioisotope lutetium-177 (177Lu) via Empiriko's proprietary RCV™ (radioactive carrier vehicle) technology. Upon binding to PSMA on the surface of cancer cells, the drug is internalized through endocytosis. EMP108 exerts its anti-tumor effect through a dual mechanism: the 177Lu isotope delivers localized beta radiation that induces DNA double-strand breaks, while the RCV carrier generates reactive singlet oxygen radicals that cause irreversible damage to cellular organelles and DNA. This synergistic approach is designed to enhance therapeutic efficacy, potentially allowing for shorter treatment durations, lower drug doses, and reduced off-target toxicity compared to conventional radioligand therapies.
