Emprumapimod (also known as ARRY-371797 and PF-07265803) is an orally bioavailable small molecule inhibitor of p38 mitogen-activated protein (MAP) kinase. Originally developed by Array BioPharma and subsequently acquired by Pfizer, the drug was designed to modulate inflammatory pathways by inhibiting the production of pro-inflammatory cytokines such as TNF-alpha and IL-1 beta. It was evaluated in a Phase 2 clinical trial for the treatment of active ankylosing spondylitis in patients who had an inadequate response to NSAIDs, but the study was terminated. Additionally, emprumapimod has been investigated for other indications, most notably dilated cardiomyopathy caused by LMNA gene mutations, as well as rheumatoid arthritis and acute dental pain.
