EMR2-CAR T-cells are an investigational chimeric antigen receptor (CAR) T-cell therapy targeting EMR2 (Adhesion G Protein-Coupled Receptor E2, also known as ADGRE2 or CD312). EMR2 is a member of the epidermal growth factor (EGF)-TM7 family of adhesion proteins, with expression primarily restricted to myeloid cells such as monocytes, macrophages, and granulocytes, making it a potential target for myeloid malignancies. The CAR construct typically utilizes a 4-1BB co-stimulatory domain (EMR2.41BB.zeta architecture) and is produced by transducing activated T-cells from peripheral blood mononuclear cells (PBMCs) with a gamma retroviral vector. It is primarily being investigated for the treatment of Acute Myeloid Leukemia (AML). Research has focused on enhancing its anti-tumor efficacy through combination with SMAC mimetics like birinapant or hypomethylating agents like decitabine.
