Overview

Emtricitabine + adefovir dipivoxil is an investigational combination of two antiviral agents, both classified as nucleoside/nucleotide reverse transcriptase inhibitors (NRTIs/ntRTIs). Emtricitabine inhibits viral reverse transcriptase by acting as a cytidine analog, while adefovir dipivoxil is an oral prodrug of adefovir, an acyclic nucleotide analog of adenosine monophosphate. Adefovir is phosphorylated intracellularly to its active form and inhibits hepatitis B virus (HBV) DNA polymerase by competing with the natural substrate and causing DNA chain termination after incorporation into viral DNA[1][5]. This combination has been studied preclinically for enhanced antiviral activity against chronic hepatitis B infection and to help manage drug resistance in HBV therapy[5]. The primary indication under investigation is chronic hepatitis B.
