**Emugrobart (GYM329, RG6237)** is a humanized IgG1κ monoclonal antibody developed by Chugai Pharmaceutical (licensed to Roche) that specifically targets and inhibits latent myostatin (GDF8, growth/differentiation factor 8), a negative regulator of muscle growth secreted primarily from muscle cells. By applying Chugai's proprietary recycling and sweeping antibody technologies, it neutralizes activated myostatin to promote muscle hypertrophy and improve conditions associated with muscle atrophy and weakness. It has orphan drug designation in the US and EU for spinal muscular atrophy (SMA) and is being evaluated in combination with risdiplam (Evrysdi) in the Phase 2/3 MANATEE trial for SMA, as monotherapy in Phase 2 for facioscapulohumeral muscular dystrophy (FSHD) and obesity, and Phase 1 for type 2 diabetes mellitus.[1][2][3][4][7]
