EN41-FPA2 is a peptide-based HIV-1 vaccine candidate developed as part of the European EuroNeut-41 consortium. The vaccine consists of a 65-amino acid synthetic peptide derived from the membrane-proximal external region (MPER) of the HIV-1 envelope glycoprotein gp41. It is designed to elicit neutralizing antibodies, specifically mucosal IgA and systemic IgG, to block HIV-1 entry at the mucosal surface and neutralize viral particles. In clinical development, EN41-FPA2 has been evaluated in Phase 1 trials using a prime-boost strategy involving intranasal priming followed by intramuscular boosting, often formulated with virosomes or other delivery systems to enhance immunogenicity.
