ENA101 is a first-in-class bispecific T cell engager (TCE) immunotherapy developed by Enara Bio. It is designed to target the novel cancer-specific "dark antigen" DARKFOX, which is a polypeptide derived from an alternative open reading frame associated with FOXM1 and is robustly expressed at the surface of solid tumor cells, with minimal expression in normal tissues. ENA101 was engineered using Enara's EnTiCE platform to achieve optimal immune synapse formation and therapeutic profile. It aims to redirect T cells to specifically attack tumor cells presenting DARKFOX, offering durable clinical benefit for a broad range of solid tumors—particularly squamous cell carcinomas, including lung, head & neck, and esophageal cancers. ENA101 is currently in IND-enabling studies and represents the first therapeutic to advance against this new class of tumor-specific antigens[3][9].
