Overview

Enadenotucirev is a tumor-selective, chimeric group B oncolytic adenovirus (Ad11p/Ad3) developed by directed evolution for the treatment of epithelial solid tumors. Developed by PsiOxus Therapeutics (now Akamis Bio), the virus is engineered to selectively replicate within malignant epithelial cells, leading to direct cell lysis via a non-apoptotic, immunogenic mechanism known as oncosis. This process releases tumor-associated antigens and inflammatory signals, stimulating an anti-tumor immune response and increasing CD8+ T-cell infiltration. Unlike many oncolytic viruses, enadenotucirev is stable in human blood and faces low pre-existing neutralizing antibody levels due to its Ad11p capsid, enabling systemic intravenous administration. It has been investigated in clinical trials for various cancers, including platinum-resistant ovarian cancer, colorectal cancer, and locally advanced rectal cancer, both as a monotherapy and in combination with chemotherapy or checkpoint inhibitors.
