**enadenotucirev + nivolumab** is a combination therapy where enadenotucirev, a tumor-selective blood-stable oncolytic adenoviral vector, is administered together with nivolumab, a PD-1 checkpoint inhibitor monoclonal antibody. Enadenotucirev selectively replicates in and lyses epithelial tumor cells, promoting immune cell infiltration and activation within the tumor microenvironment. Nivolumab blocks the interaction between PD-1 and its ligands, PD-L1/PD-L2, thereby enhancing anti-tumor immune responses by reactivating cytotoxic T lymphocytes. This combination is being investigated as an approach to overcome resistance to PD-1 blockade, particularly in advanced/metastatic epithelial cancers, including colorectal cancer (CRC) and squamous cell carcinoma of the head and neck (SCCHN), with a focus on microsatellite stable/microsatellite instability-low patients. Early-phase clinical trials have shown manageable tolerability, increased CD8+ T-cell infiltration, and encouraging overall survival, although response rates remain modest[1][3][5][6][7][8].
