EnanDIM (Enantiomeric DNA-based ImmunoModulator) is a novel family of Toll-like receptor 9 (TLR9) agonists developed by Mologen AG for cancer immunotherapy. These molecules are synthetic, DNA-based oligonucleotides containing multiple non-methylated CG-motifs. A distinguishing structural feature is the replacement of D-deoxyribose nucleotides at the 3'-ends with nuclease-resistant L-deoxyribose nucleotides, which protects the molecule from degradation without the need for phosphorothioate modifications. By activating TLR9, EnanDIM triggers a broad immune response, including the activation of dendritic cells and natural killer cells, leading to anti-tumor activity. Preclinical studies demonstrated efficacy in various murine tumor models, including colon carcinoma, melanoma, lymphoma, and breast cancer. A Phase 1 clinical trial was initiated for patients with advanced solid tumors, but development was halted following the insolvency of Mologen AG.
