Enapotamab is a human monoclonal antibody (IgG1-kappa) that targets AXL, a member of the TAM (Tyro3, AXL, Mer) family of receptor tyrosine kinases. AXL is frequently overexpressed in various malignancies, including multiple myeloma, where it contributes to tumor cell survival, epithelial-to-mesenchymal transition (EMT), and resistance to therapy. While originally developed as the antibody component of the antibody-drug conjugate (ADC) enapotamab vedotin (HuMax-AXL-ADC), enapotamab is being investigated as a standalone therapeutic or in combination regimens. In the context of relapsed or refractory multiple myeloma, it is being studied in combination with mezigdomide (a CELMoD agent) to evaluate potential synergistic effects in overcoming drug resistance and improving clinical outcomes.
