Enavatuzumab (PDL192; ABT-361) is a first-in-class humanized IgG1 monoclonal antibody that targets the Tumor necrosis factor-like weak inducer of apoptosis receptor (TWEAK receptor, also known as TweakR or TNFRSF12A), which is the receptor for TNF-like weak inducer of apoptosis (TWEAK). It acts as a moderate agonist of TweakR and mediates anti-tumor activity through both direct signaling via TweakR and by inducing antibody-dependent cellular cytotoxicity (ADCC). Unlike its natural ligand TWEAK, enavatuzumab does not stimulate cytokine expression strongly nor promote angiogenesis. The drug has demonstrated antitumor effects in preclinical models and was evaluated in phase 1 clinical trials for advanced solid tumors and breast cancer. Development was led by Abbott/AbbVie[1][3][5][7].
